Phenotyping Approach:

The SLAD plates used to screen PSH were washed in order to analyze IG. After imaging PSH phenotypes, the SLAD plates were gently washed using DI water to remove the yeast colony remaining on the surface level of the agar. This “wash step” enables the isolation of yeast cells penetrating the solid surface invasively. The washed SLAD plates were imaged using an Epson Expression 11000 XL scanner.

A python pipeline was used to process pre-washed and post-washed images of the SLAD 2x plates by applying a threshold to the images to divide the pixels in an image into two clusters: (1) colony, (2) background. IG was quantified by dividing the number of colony pixels in the post-wash processed image (d) by the number of colony pixels in the pre-wash processed image (c).
